This antibody is provided in a ready-to-use formulation.
Recommended positive control tissue: Rhabdomyosarcoma (not limited to this tissue type).
Intended for professional laboratory use in detecting myogenin protein in formalin-fixed, paraffin-embedded tissues using manual qualitative IHC.
Used after primary tumor diagnosis by conventional histopathology.
Myogenin belongs to the MyoD family of myogenic transcription factors (including MyoD, Myf-5, and MRF4). These factors are specific to skeletal muscle and activate myogenesis by binding to enhancer sequences of muscle-specific genes.
Anti-myogenin labels nuclei of myoblasts and is expressed in rhabdomyosarcoma and some leiomyosarcomas. Positive nuclear staining may also occur in Wilms' tumor.
Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended.
IHC staining visualizes antigens through sequential application of:
- Primary antibody
- Secondary (link) antibody
- Enzyme complex
- Chromogenic substrate
The enzymatic reaction produces a visible signal at the antigen site, interpreted under a light microscope for differential diagnosis.
A positive tissue control should be included in each staining procedure.
- Tissue should contain both positive and negative staining regions.
- External positive controls: freshly fixed and processed specimens.
- Weakly positive tissues are ideal for detecting subtle reagent degradation.
Negative controls (internal or external) verify absence of specific staining and help identify background noise.
If specific staining appears in negative controls, patient specimen results must be considered invalid.

It belongs to the helix-loop-helix protein family and can induce myogenesis in various cell types.
