Target Information
This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant).
These chemokines belong to the CXC chemokine family, characterized by a single amino acid separating the first two conserved cysteine residues.
Binding of chemokines to this receptor triggers leukocyte trafficking responses such as integrin activation, cytoskeletal rearrangement, and chemotaxis.
The receptor is coupled to Gi-type G proteins and participates in rapid, transient signaling events that are desensitized via phosphorylation-dependent internalization.
It is expressed prominently in effector/memory T cells and inflamed tissues, suggesting a role in inflammatory cell recruitment.
CXCR3 is therefore a target for small-molecule antagonist development for inflammatory disease treatment.
Multiple transcript variants encoding different isoforms have been identified.
