Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. Antibody DNA sequences from immunoreactive rabbits are cloned and screened to select optimal production candidates.

Target Information

HLA-DQA1 binds peptides derived from antigens that access the endocytic route of antigen-presenting cells (APCs) and presents them on the cell surface for recognition by CD4 T-cells. The peptide binding cleft accommodates peptides of 10–30 residues.
Peptides presented by MHC class II molecules are generated by degradation of proteins processed by lysosomal proteases. Exogenous antigens endocytosed by APCs are presented via MHC II molecules, the exogenous antigen presentation pathway.
Endogenous peptides can also be presented via autophagy, where autophagosomes fuse with MHC class II loading compartments.
In addition to APCs, epithelial cells of the gastrointestinal tract express MHC class II molecules and CD74, functioning as APCs.
For antigen presentation, three MHC class II heterodimers associate with a CD74 trimer in the ER to form a heterononamer. CD74 is sequentially degraded, leaving CLIP fragments, which are removed by HLA-DM to allow high-affinity peptide binding.
HLA-DM stabilizes MHC II molecules until antigenic peptides are bound. In B-cells, this interaction is regulated by HLA-DO. Lysosomal acidification enhances proteolysis and peptide loading efficiency.

HLA and MHC antibodies are crucial in immunopeptidomics, aiding identification and characterization of neoantigens through LC-MS/MS analysis.
