Target Information
Alpha 2-Macroglobulin (A2M) is an approximately 720-kDa homotetrameric glycoprotein composed of four identical ~180 kDa subunits. A2M shares structural similarities with other alpha-macroglobulins such as complement components C3 and C4 and pregnancy zone protein (PZP), exhibiting a strong binding capacity for various ligands. This enables alpha-macroglobulins to act as humoral defense barriers against foreign peptides in plasma. A2M captures a wide range of proteinases, functioning as a panprotease inhibitor. In the brain of Alzheimer’s disease patients, A2M has been localized to diffuse amyloid plaques, suggesting its significant role in AD pathophysiology.
