Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It is a ligand-activated transcription factor that binds active testosterone (T) and dihydrotestosterone (DHT). Upon hormone binding, the receptor translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

AR signaling is essential for male reproductive organ development and function and also affects nonreproductive tissues such as muscle, hair follicles, and brain. The receptor is a phosphoprotein that regulates MAP kinase activity, and its phosphorylation state correlates with MEK1/2 pathway inhibition. Dysregulation of AR signaling is associated with diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in AR can cause complete androgen insensitivity (CAIS).
