LRP6 is essential for the Wnt/β-catenin signaling pathway, acting as a coreceptor with Frizzled for Wnt.

The interaction with DKK1 blocks LRP6-mediated Wnt/β-catenin signaling via Kremen proteins-mediated endocytosis.
