Target Information
BRCA1-associated protein-1 (BAP1)는 BRCA1의 RING finger 도메인과 상호작용하는 단백질로, 인간 단백질의 N-말단 240개 아미노산은 ubiquitin C-terminal hydrolases (UCHs)와 상동성을 보입니다. BAP1은 산성 영역, 고전하 C-말단 영역, 두 개의 핵 내 위치 신호(NLS)를 포함하며, BRCA1과 유사한 세포핵 내 분포를 보입니다.
BAP1은 BRCA1 매개 유방암 세포 성장 억제를 강화하며, 모든 인간 조직에서 4-kb mRNA로 발현되고, 고환에서는 4.8-kb 전사체가 특이적으로 발현됩니다. 또한 BAP1과 BRCA1은 생쥐 유방 발달 및 재구성 과정에서 공동 발현됩니다.
BAP1 유전자는 염색체 3p21.3에 위치하며, 이 영역은 유방암 및 폐암에서 종종 결실되는 부위로 알려져 있습니다. BAP1의 유전자 내 재배열 및 결실은 폐암 세포주에서 관찰되며, BAP1은 BRCA1 성장 조절 경로에서 종양 억제 유전자로 기능할 가능성이 제시되었습니다.
