Description:
The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), the low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa.
CD16 includes two genes: CD16A (FcγRIIIA) expressed in macrophages and NK cells, and CD16B (FcγRIIIB) expressed in neutrophils. CD16 binds IgG immune complexes, showing preferential binding to IgG1 and IgG3, and minimal binding to IgG2 and IgG4.
Upon IgG binding, CD16 isoforms trigger signal transduction leading to ADCC, phagocytosis, degranulation, and proliferation.

Applications Reported:
This CB16 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test. A test is defined as staining a cell sample in 100 µL final volume. Cell number can range from 10⁵–10⁸ cells/test.

Fluorophore Information:
APC-eFluor 780 emits at 780 nm and is excited by the Red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and compensation.

Excitation / Emission / Laser:
633–647 nm excitation; 780 nm emission; Red laser.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD16 encodes the Fc receptor for IgG, involved in immune complex clearance and antibody-dependent cellular cytotoxicity.
The FCGR3A gene is expressed on NK cells as a transmembrane glycoprotein, while FCGR3B is expressed on neutrophils via PI linkage.
Mutations in FCGR3A are linked to immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and neonatal neutropenia.
Alternative splicing leads to multiple isoforms.
