Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays an essential role in T cell signal transduction, differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules. In contrast, CD4 interacts with MHC class II molecules. CD8α is critical for MHC-I binding and is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers. Engagement of MHC-I/peptide complexes on antigen-presenting cells recruits Lck kinase, initiating lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are key in eliminating pathogens and tumor cells. Differentiation of naive CD8⁺ T cells into CTLs is promoted by IL-2, IL-12, and TGF-β1.
