Target Information
Tau is a neuronal microtubule-associated protein predominantly found on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules, while the N-terminus binds neural plasma membrane components, functioning as a linker between them. Axonal polarity is determined by TAU/MAPT localization within the neuronal cell body domain defined by the centrosome.
Short isoforms contribute to cytoskeletal plasticity, whereas longer isoforms aid in stabilization. Hyper-phosphorylated Tau forms paired helical filaments (PHF), the main component of neurofibrillary tangles in Alzheimer’s disease (AD) brains. This modification impairs Tau’s microtubule-binding ability, leading to destabilization and neuronal degeneration.
Tau phosphorylation is mediated by several kinases, including GSK-3β, PKA, cdk5, and casein kinase II. Hyper-phosphorylated Tau is also observed in other neurodegenerative conditions such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
