Target Information

Gab1 is a 115 kDa multiple docking protein essential for cellular growth, transformation, and apoptosis.
It can be phosphorylated by multiple receptor tyrosine kinases (RTKs), including:

• Insulin receptor (IR)
• Platelet-derived growth factor receptor beta (PDGFRβ)
• Hepatocyte growth factor/scatter factor receptor (HGFR/SFR or c-Met)
• Epidermal growth factor receptor (EGFR)

Gab1 undergoes tyrosine phosphorylation at least at 16 sites, serving as binding sites for PI3K, Grb2, PLCγ1, Nck, and SHP2.
Phosphorylation on tyrosines 627 and 659 is critical for SHP2 binding and activation of the ERK/MAPK pathway in response to EGF.
