PGA5 (Pepsinogen 5, group I / Pepsinogen A)
Pepsinogens are inactive precursors of pepsin, the major acid protease found in the stomach. Pepsin is a single polypeptide chain derived from pepsinogen by removal of a 41-amino-acid segment from the N-terminus. It is synthesized in the stomach lining, where hydrochloric acid converts it into the active enzyme and maintains the optimal pH (1–3) for its function.
Pepsin effectively cleaves peptide bonds involving aromatic amino acids and shows broad specificity, preferring bonds with phenylalanine and leucine. PGA5 belongs to the A1 subfamily within the pepsin family and is the predominant endopeptidase in vertebrate gastric juice. It is inhibited by ovUS-1, a uterine serpin.
