Invitrogen™ LentiArray™ Human GPCR CRISPR Library
The Invitrogen™ LentiArray™ Human GPCR Library targets 446 genes with up to 4 gRNAs per gene target (pooled in a single well), for a total of 1,784 gRNAs.
Libraries are delivered as 100 μL of ready-to-use lentiviral particles per gene target with an average titer of 1×10⁸ TU/mL (functional titer determined by antibiotic resistance).
Overview
G protein-coupled receptors (GPCRs) regulate a wide range of physiological and disease processes. Nearly 30% of FDA-approved drugs target GPCRs, making them one of the most druggable gene classes.
The gene targets in this library were selected using up-to-date genome databases, including NCBI RefSeq, Gene Ontology Consortium (GO), and HUGO Gene Nomenclature Committee (HGNC).

LentiArray CRISPR libraries use Thermo Fisher Scientific’s proprietary CRISPR gRNA design algorithm, incorporating the latest research and in-house experience to maximize editing efficiency and specificity.
Each gene target includes up to four high-quality gRNAs designed to knock out all known isoforms, ensuring efficient knockout across diverse cell types.
Flexible Experimental Design
The library employs a two-vector system, expressing Cas9 nuclease and gRNA on separate lentiviral constructs.
This allows users to control when and how genome-editing tools are delivered. The LentiArray Cas9 lentivirus expresses a human codon-optimized Cas9 with a blasticidin-resistance gene under the EF-1α (EFS) promoter.
Users can:
- Co-infect cells with both Cas9 and gRNA lentiviruses for immediate editing
- Establish stable Cas9-expressing cell lines using only the Cas9 lentivirus
The gRNA lentiviruses do not contain fluorescent markers, allowing the use of reporters of any wavelength and enabling multiplexed experiments.
For challenging cell lines, puromycin resistance allows enrichment of edited cells and stronger phenotypes.
