Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 protein tyrosine phosphatases, and these motifs also mediate ubiquitin-dependent receptor removal from the cell surface.
CD33 is expressed on cells of the myelomonocytic lineage, binds sialic acid residues in N- and O-glycans, and serves as a therapeutic target for acute myeloid leukemia. It is found on granulocyte and macrophage precursors in bone marrow but not on pluripotent stem cells. CD33 expression on peripheral monocytes makes it a useful marker for distinguishing myelogenous leukemia from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
