Target Information

MAP3K14 (NIK) is structurally related to MAP3Ks. MAP3K14 phosphorylates and activates CHUK (IKKα) and IKBKB (IKKβ), which in turn phosphorylate IκB-α, releasing NF-κB and leading to up-regulation of inflammatory response genes. This kinase binds to TRAF2 and stimulates NF-κB activity. It shares sequence similarity with several other MAPKK kinases and participates in an NF-κB-inducing signaling cascade common to receptors of the TNF/NGF family and the interleukin-1 type-I receptor.
