Target Information

Various biochemical, physiological, and behavioral processes display circadian rhythms controlled by an internal biological clock. The central gears driving this clock appear to be composed of an autoregulatory transcription/post-translation-based feedback loop.
Cryptochrome 1 (CRY1) and 2 (CRY2) are DNA-binding flavoproteins that share homology with blue-light receptors and photolyases.
In Drosophila, CRY acts as a photoreceptor for the circadian clock, binding to TIM in a light-dependent manner.
Mammalian CRY1 and CRY2 interact with circadian genes CLOCK, BMAL1, PER1, PER2, and TIM in a light-independent fashion, regulating Per1 transcriptional cycling.
Mutant mice lacking Cry1 or Cry2 exhibit altered locomotor activity periodicity, indicating that both proteins are essential for synchronizing circadian phases.
A balance between Cry1 and Cry2 is critical for proper clock function; double-mutant mice in darkness show arrhythmicity, signifying loss of internal circadian rhythm.
