Product Specific Information
The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as either a heterodimer (CD8αβ) or homodimer (CD8αα). CD8a is found on thymocytes, mature T cells, and NK cells. It binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a critical role in T-cell development and activation.
Preliminary testing indicates OKT8 does not compete with clones RPA-T8 or HIT8a for binding to human peripheral blood leukocytes, suggesting distinct epitopes.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometry on normal human peripheral blood cells at ≤0.06 µg/test. A test is defined as staining a cell sample (10⁵–10⁸ cells) in 100 µL final volume. Optimal performance requires titration.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin family. It functions as a co-receptor for MHC class I molecules and is predominantly expressed on cytotoxic T cells. CD8 exists as CD8αβ heterodimers or CD8αα homodimers on T cells, NK cells, monocytes, and dendritic cells.
Binding of MHC-I/peptide complexes to CD8 triggers Lck recruitment, leading to lymphokine production, motility, and CTL activation. Activated CTLs play a crucial role in pathogen and tumor cell clearance. Differentiation into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
