Target Information
CHEK1 (CHK1) is a serine/threonine protein kinase involved in preventing replication when DNA integrity is compromised.
CHK1 plays a key role in the DNA damage-induced checkpoint network and ensures genomic integrity under genotoxic stress.
When CHK1 inhibition abrogates the G2 or S checkpoint, p53-deficient cancer cells undergo mitotic catastrophe and apoptosis, while normal cells remain arrested in the G1 phase.
Thus, CHK1 inhibitors can enhance the efficacy of DNA-damaging agents in cancer cells, making CHK1 an attractive therapeutic target, especially since approximately 50% of human cancers are p53-deficient.
