Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding mature erythrocytes and platelets. It plays a critical role in lymphocyte activation through its tyrosine phosphatase activity.

• Applications Tested: Tested on mouse bone marrow cells at ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)

Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended detection filter: 660/20 bandpass.

When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.
Protect from light due to photo-induced oxidation sensitivity.

Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, constituting about 10% of lymphocyte surface proteins. It is absent in non-hematopoietic cells and tissues.

CD45 regulates Src-family kinases, T- and B-cell receptor signaling, and cytokine receptor signaling by suppressing JAK kinases. It plays a crucial role in lymphocyte development, antigen signaling, integrin-mediated pathways, and apoptosis regulation.
Different isoforms (e.g., CD45RA) arise from alternative splicing and vary by cell type and physiological state.
