Target Information
DNA methylation, or the addition of methyl groups to cytosine bases in the dinucleotide CpG, is crucial for proper development and gene expression regulation. Methylation and demethylation are enzymatic processes that determine methylation patterns. The demethylation enzyme, a mammalian protein associated with a methyl-CpG-binding domain (MBD), exhibits demethylase activity capable of reverting methylated cytosine bases to cytosine by cleaving the methyl group as methanol.
Proteins such as MeCP2 and MBD1 repress transcription by binding specifically to methylated DNA. MBD2 and MBD4 (also known as MED1) colocalize with heavily methylated satellite DNA and are believed to mediate the biological functions of methylation signals. Unlike others, MBD3 does not bind methylated DNA in vivo or in vitro. MBD1, MBD2, MBD3, and MBD4 are expressed in somatic tissues, while MBD1 and MBD2 expression is reduced or absent in embryonic stem cells, which lack MeCP1 activity. MBD4 shares homology with bacterial base excision repair DNA N-glycosylases/lyases and is found mutated in some microsatellite unstable tumors.
