Target Information
Mitochondrial inner membrane calcium uniporter (MCU) mediates calcium uptake into mitochondria and forms the pore-forming subunit of the uniporter complex. Its activity is regulated by MICU1 and MICU2: inhibited at low Ca²⁺ levels and activated at higher levels. MCU plays a central role in mitochondrial calcium homeostasis, influencing cellular physiology, energy metabolism, and apoptosis regulation.
It contributes to buffering systolic calcium levels in cardiomyocytes and supports the “fight-or-flight” response by enabling rapid mitochondrial calcium uptake in pacemaker cells. MCU is also implicated in mitochondrial permeability transition during ischemia-reperfusion injury, glucose-dependent insulin secretion in pancreatic β-cells, muscle size regulation in skeletal muscle, and synaptic vesicle endocytosis in the nervous system. Additionally, it participates in antigen processing and presentation.
