Product Specific Information

MA1-216 detects thyroid hormone receptor (TR) beta-1 in human, mouse, rat, and transformed bacterial cells. This antibody is specific for the TR beta-1 isoform.
MA1-216 has been successfully used in Western blot, immunocytochemistry, immunoprecipitation, and gel shift procedures. By Western blot, this antibody detects 52 and 55 kDa proteins representing recombinant TR expressed in E. coli, corresponding to two different translational start points.
Immunocytochemical staining in GH3 cells shows nuclear localization, and at high expression levels, additional cytoplasmic staining. Immunoprecipitation and gel super shift experiments confirm that MA1-216 reacts with unliganded, liganded, and DNA-binding forms of the receptor.
The immunogen is purified human TR beta-1 overexpressed in E. coli, recognizing an epitope in the A/B domain (amino acids 1–101).

Target Information

Thyroid hormone receptors (TR), also known as c-erbA-2, are ligand-dependent intracellular proteins that regulate transcription by binding to hormone response elements following activation by thyroid hormones.
Through TR interactions, thyroid hormones influence metabolism, growth, and development by regulating genes such as growth hormone, malic enzyme, and hepatic proteins.
There are two main forms of TR:

  • TR alpha (chromosome 17)
  • TR beta (chromosome 3)

Each form has two isoforms: TR alpha-1/TRv alpha-2 and TR beta-1/TR beta-2. TR alpha-1 and TRv alpha-2 share identical sequences up to amino acid 370, after which they diverge. TRv alpha-2 does not bind T3 and acts as a strong negative regulator.
The gene encoding thyroid hormone receptor beta-1 is NR1A2.
