Target Information
The G-protein-coupled receptor (GPCR) superfamily comprises approximately 600–1,000 members and represents the largest class of molecular targets with significant therapeutic potential.
GPR26 is a brain-specific orphan GPCR exhibiting constitutive activity mediated by G(s)-alpha protein, which stimulates adenylate cyclase and elevates intracellular cAMP.
In the central nervous system, GPR26 shows highest expression in the amygdala, hippocampus, and thalamus, with weaker expression in the testis.
GPR26 is down-regulated in glioblastoma. Depletion of GPR26 increases fat storage in C. elegans, and deficiency in the hypothalamus is linked to obesity susceptibility in mice, along with metabolic complications such as glucose intolerance, hyperinsulinemia, and dyslipidemia.
