Target Information

RIPK2 is a serine/threonine kinase involved in cell activation, proliferation, differentiation, and apoptosis. Upon stimulation by bacterial peptidoglycans, NOD1 and NOD2 are activated, oligomerize, and recruit RIPK2 through CARD-CARD domains.
RIPK2 contributes to the tyrosine phosphorylation of ARHGEF2 through Src kinase, leading to NF-κB activation by NOD2. Once recruited, RIPK2 autophosphorylates and undergoes Lys-63-linked polyubiquitination by E3 ligases (XIAP, BIRC2, BIRC3).
The polyubiquitinated protein mediates recruitment of MAP3K7/TAK1 to IKBKG/NEMO, inducing Lys-63-linked polyubiquitination of IKBKG/NEMO and subsequent activation of IKBKB/IKKB. This results in NF-κB release and nuclear translocation, activating genes related to immune response, growth control, and apoptosis protection.
RIPK2 also plays a role in T-cell receptor (TCR) engagement by promoting BCL10 phosphorylation and subsequent NF-κB activation.
