Target Information
Lipolytic enzymes are essential for mobilizing fatty acids from triglyceride stores in adipose tissue. Dysfunctional lipolysis affects energy homeostasis and may contribute to obesity and insulin resistance. Hormone-sensitive lipase (HSL) was previously known as the main enzyme for triglyceride hydrolysis, but adipose triglyceride lipase (ATGL) has been identified as initiating triglyceride breakdown. ATGL is highly expressed in adipose tissue of mice and humans, specifically hydrolyzing triacylglycerol and associating with lipid droplets. Inhibition of ATGL decreases total adipose acyl-hydrolase activity, showing that ATGL and HSL work together to catabolize stored triglycerides in mammals.
