Product Specific Information

The monoclonal antibody B6H12 reacts with CD47, also known as integrin-associated protein (IAP) and neurophilin.
CD47 is a glycosylated five-transmembrane protein involved in adhesion through interactions with SIRP (signal regulator protein) and integrins (CD51/CD61 and CD41/CD61).
These interactions mediate bi-directional signaling, influencing neural synaptic activity and macrophage phagocytic regulation.
CD47 also acts as a receptor for thrombospondin, mediating T cell activation or apoptosis depending on ligand levels.
Stimulation of CD47 by monoclonal antibodies can induce CD4⁺CD25⁻ suppressive activity and upregulate Foxp3 expression.

Expression is found in most hematopoietic cells (T, B, monocytes, platelets, erythrocytes) and non-hematopoietic cells.
This antibody has been reported to have neutralizing activity.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (1.0 µg)/test.
A test corresponds to staining in 100 µL final volume. Cell number may vary (10⁵–10⁸ cells/test).

Target Information

CD47 (integrin-associated protein, IAP) is a transmembrane glycoprotein that interacts with integrins and regulates their functions.
Engagement with ligands or counter receptors modulates signaling pathways, including G protein activation and calcium regulation during cell adhesion.
It serves as a receptor for thrombospondin’s C-terminal domain, influencing membrane transport and signal transduction.
CD47 interacts with CD172a (SIRPα) on macrophages to inhibit phagocytosis of CD47⁺ cells.
It also regulates cell migration, B cell adhesion, T cell activation, and neuronal development, being abundant in synapse-rich brain and retinal regions.
