Target Information

RYBP is a component of a Polycomb group (PcG) multiprotein PRC1-like complex, which maintains the transcriptionally repressive state of many genes, including Hox genes, throughout development. This complex acts via chromatin remodeling and histone modification, mediating monoubiquitination of histone H2A at Lys-119, leading to heritable changes in chromatin expressibility.
RYBP is involved in monoubiquitination of histone H2A Lys-119 on the X chromosome and is required for normal silencing of one copy of the X chromosome in XX females. It may stimulate histone ubiquitination by recruiting the complex to target sites and inhibit ubiquitination and degradation of TP53, thus regulating transcription of TP53 target genes.
Additionally, RYBP may regulate ubiquitin-mediated proteasomal degradation of proteins such as FANK1 to control apoptosis and act as a transcriptional repressor of E4TF1 activity. It may bind DNA and play a role in repression of tumor growth and metastasis in breast cancer by down-regulating SRRM3.
