Target Information

Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase subfamily that includes EGFR, ERBB2, and ERBB3. ERBB4 binds and is activated by neuregulins and other factors, inducing various cellular responses such as mitogenesis and differentiation.
It acts as a receptor for heregulin and mediates HGL-stimulated tyrosine phosphorylation. Like other protein kinases, ErbB4 regulates signal transduction in eukaryotic cells, influencing metabolism, transcription, cell cycle, cytoskeletal rearrangement, cell movement, apoptosis, and differentiation.
ErbB4 is highly expressed in several breast carcinoma cell lines (T47-D, MDA-MB-453, BT-474, H3396) and in normal skeletal muscle, heart, pituitary, brain, and cerebellum. Intermediate expression is observed in MCF-7, MDA-MB-330, and MDA-MB-361, while low or absent expression is found in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3. Elevated ErbB4 levels in certain tumor cell lines suggest a potential role in malignancy.
