Target Information
Cell cycle progression is subject to arrest at G1 and G2 checkpoints in response to DNA damage, allowing time for DNA repair before entering S and M phases. The p53 tumor suppressor regulates one such G1 checkpoint and upregulates GADD45 and p21 expression. GADD45 binds Cdks and PCNA, stimulating DNA excision repair and inhibiting S phase entry. GADD45 may serve as a link between the p53-dependent cell cycle checkpoint and DNA repair. GADD45-like proteins, GADD45 beta and GADD45 gamma, are induced by environmental stresses and thought to activate p38/JNK signaling via MEKK4 activation.
