Target Information

CRTH2 (GPR44) is a chemoattractant receptor-homologous molecule expressed on Th2 cells and acts as a receptor for prostaglandin D2 (PGD2). It mediates chemotaxis of Th2 cells and eosinophils toward PGD2, playing a key role in allergic responses.
CRTH2-deficient mice show enhanced eosinophil recruitment and increased IL-5 and IL-3 production, suggesting its role in restricting eosinophilia and allergic responses.
CRTH2 is a member of the G protein-coupled leukocyte chemoattractant receptor family, selectively expressed in Th2 lineage cells. It is found in the central nervous system, peripheral tissues, and activated T helper-2 cells.
Two transcripts have been identified:

• 3.5 kb transcript: expressed in thalamus, frontal cortex, pons, hippocampus
• 3.4 kb transcript: expressed in fetal liver, leukocytes, thymus

CRTH2 expression is associated with activated Th2 cells, suggesting its pivotal role in Th2-type immune reactions. Single nucleotide polymorphisms in the 3′ UTR of the CRTH2 gene have been linked to asthma susceptibility.
