Target Information
DNA-directed RNA polymerase II subunit RPB1 (POLR1A) is a DNA-dependent RNA polymerase that catalyzes the transcription of DNA into RNA using ribonucleoside triphosphates as substrates. POLR1A is the largest subunit and a catalytic component of RNA polymerase II, which synthesizes mRNA precursors and many functional non-coding RNAs. It forms the polymerase active center with the second largest subunit and is part of the core element that includes the central cleft, clamp, and jaws that interact with DNA.
During transcription initiation, the single-stranded DNA template is positioned within the active site cleft of Pol II. The RPB1 subunit contributes a bridging helix that promotes translocation of Pol II by acting as a ratchet mechanism. Transcription elongation is modulated by phosphorylation of the C-terminal domain (CTD) of RPB1, which serves as a platform for assembly of regulatory factors involved in initiation, elongation, termination, and mRNA processing. The balance between methylation and acetylation of CTD lysines is critical for gene expression regulation.
