Product Specific Information

The A20 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.1-expressing strains such as SJL/J, DA, STS/A, and RIII. CD45.1 is expressed on all leukocytes in these strains.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Optimize titration for best performance.
• Fluorochrome: Super Bright 702 (excited by 405 nm violet laser, emits at 702 nm; use 710/50 bandpass filter).
• Light Sensitivity: Protect from light due to photo-induced oxidation.
• Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333).
• Buffer Recommendation: When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401) to minimize non-specific polymer interactions.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins.
It regulates Src-family kinases, influences lymphocyte development and antigen signaling, and exists in multiple isoforms due to alternative splicing.
CD45 isoforms differ in extracellular domains and expression patterns depending on cell type and physiological state.
It also contributes to cell survival and apoptosis regulation via integrin-mediated signaling and DNA fragmentation pathways.
CD45RA is one isoform with restricted expression among lymphoid subtypes.
