Target Information

Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene.
It is a key structural component of the extracellular matrix in cartilage, providing resistance to compression. The human protein consists of 2316 amino acids (~250 kDa) and exists in multiple isoforms due to alternative splicing.

Aggrecan has a bottlebrush-like structure, with chondroitin sulfate and keratan sulfate chains attached to a central protein core. It contains three interglobular domains (G1, G2, and G3) that contribute to aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis.

Fragments of Aggrecan are released into synovial fluid during osteoarthritis progression due to cleavage by aggrecanase and MMPs. Antibodies such as BC-3 and BC-4 can detect neo-epitopes of Aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
