Target Information
ABL1 원발암유전자는 세포 분화, 세포 분열, 세포 부착 및 스트레스 반응 과정에 관여하는 세포질 및 핵 단백질 티로신 키나아제를 암호화합니다.
c-Abl 단백질의 활성은 SH3 도메인에 의해 음성 조절되며, SH3 도메인이 결실되면 c-Abl은 종양유전자로 전환됩니다.
ABL1 티로신 키나아제의 DNA 결합 활성은 CDC2 매개 인산화에 의해 조절되어 세포 주기 기능을 시사합니다.
만성 골수성 백혈병 및 일부 급성 림프구성 백혈병에서 c-Abl 원발암유전자는 (9;22) 염색체 전위로 인해 Philadelphia 염색체를 형성하며, 이는 9번 염색체의 c-Abl 서열이 22번 염색체의 Bcr 유전자와 융합된 결과입니다.
c-Abl 종양유전자는 Abelson murine leukemia virus (A-MuLV)의 바이러스 변형 유전자로 처음 확인되었습니다.
또한 c-ABL은 DNA 손상이 복구 불가능할 정도로 심할 때 세포자멸 경로를 활성화하여 DNA 복구를 조절할 수 있습니다.
