This antibody recognizes the chicken homologue of human CD184, also known as CXCR4 (C-X-C Receptor type 4) and fusin. CXCR4 is a receptor for the chemokine stromal-derived factor-1 (SDF-1), also known as CXCL12. Bursa cells may be used as a positive control for CXCR4 in flow cytometry.

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a member of the G-protein-coupled chemokine receptor family with seven membrane-spanning domains and functions as a co-receptor for X4 HIV-1 entry into CD4+ cells. It also acts as a co-receptor for HIV-2 binding to CD4 through envelope glycoprotein gp120, promoting Env-mediated fusion. CXCR4 serves as a receptor for CXCL12/SDF-1, mediating intracellular calcium signaling and MAPK1/MAPK3 activation. It also binds extracellular ubiquitin, influencing calcium and cAMP levels. CXCR4 is involved in hematopoiesis, cardiac septum formation, and vascular development in the gastrointestinal tract. In the CNS, it contributes to neuronal survival and cerebellar development. CXCR4 also mediates LPS-induced inflammatory responses, including TNF secretion by monocytes. Antibodies to CXCR4 can block HIV-1 and HIV-2 fusion and infection of human target cells. The amino-terminal domain and the second extracellular loop of CXCR4 serve as HIV binding sites.
