Target Information

IGF1R (Insulin-like Growth Factor 1 Receptor) is a receptor tyrosine kinase activated by IGF-1 and IGF-2, implicated in various cancers. The protein forms a tetramer with two alpha and two beta subunits. The alpha-subunits bind IGF-1 with higher affinity than IGF-2, inducing conformational change and autophosphorylation at positions 1134, 1135, and 1136. Activated IGF1R phosphorylates substrate proteins, triggering RAS, RAF, and MAPK pathways (ERK, p38, JNK), leading to gene transcription and cell proliferation.
