Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is implicated in human retinal disease. It regulates cell survival, migration, differentiation, and efferocytosis (phagocytosis of apoptotic cells). Ligand binding induces autophosphorylation of MERTK, creating docking sites for downstream signaling molecules such as GRB2 and PLCG2. Activation leads to phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1. MERTK signaling contributes to macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and engulfment. In RPE, it regulates rod outer segment fragment phagocytosis and inhibits Toll-like receptor (TLR)-mediated innate immune responses via STAT1 activation, inducing SOCS1 and SOCS3 production.
