Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is associated with retinal diseases. It regulates processes such as cell survival, migration, differentiation, and efferocytosis (phagocytosis of apoptotic cells). Ligand binding induces autophosphorylation on the intracellular domain, creating docking sites for downstream signaling molecules such as GRB2 and PLCG2. Activation leads to phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1. MERTK signaling contributes to macrophage clearance, platelet aggregation, cytoskeleton reorganization, and inhibition of Toll-like receptor-mediated innate immune responses via STAT1 activation and SOCS1/SOCS3 production. Functions as a regulator of rod outer segment phagocytosis in RPE.
