Target Information
USP7 (HAUSP)는 기질 단백질로부터 유비퀴틴을 절단하는 탈유비퀴틴화 효소입니다. 유비퀴틴화는 일반적으로 세포 단백질의 안정성과 분해에 관련되어 있으며, HAUSP의 활성은 기질 단백질을 안정화시킵니다. 특히 HAUSP는 종양억제 단백질 p53의 E3 유비퀴틴 리가아제인 Mdm2의 직접적인 길항제로 알려져 있습니다. 정상 상태에서는 Mdm2에 의해 p53이 유비퀴틴화되어 분해되지만, HAUSP는 p53을 탈유비퀴틴화하여 Mdm2 매개 분해로부터 보호함으로써 종양 억제 기능을 나타낼 수 있습니다. 또한 Myc 및 E1A와 같은 온코진에 의해 활성화된 p53 경로에서도 HAUSP가 세포 보호 역할을 수행할 가능성이 제시되고 있습니다.
