Target Information
G protein–coupled inwardly rectifying potassium channels (Kir3.1–Kir3.4) are coupled to multiple neurotransmitter receptors in the brain and are abundantly expressed in the olfactory bulb, hippocampus, neocortex, dentate gyrus, cerebellar cortex, and thalamus.
Also known as GIRK channels, Kir3 proteins localize to the soma, dendrites, and axons of neurons. Gβγ subunits from G protein heterotrimers bind directly to Kir3 cytoplasmic domains, modulating channel activity. Gβ3- and Gβ4-containing dimers enhance K⁺ current, while Gβ5-containing dimers inhibit it. Kir3 activity is also inhibited by tyrosine phosphorylation, which can be induced by brain-derived neurotrophic factor (BDNF) through receptor tyrosine kinase B activation.
