Target Information
Leukemia inhibitory factor (LIF) is a 20 kDa protein belonging to the IL-6 receptor family. It binds to a heterodimeric membrane receptor composed of the LIF-specific subunit gp190 (LIFR) and gp130, which is shared among IL-6 family members. LIF is expressed in various tissues such as thymus, lung, and neuronal tissue. It can be up-regulated by pro-inflammatory cytokines (e.g., TNFα, IL-17) and is elevated in rheumatoid arthritis, neural injury, systemic inflammation, and tuberculosis. LIF inhibits embryonic stem cell differentiation in mice and supports stem cell self-renewal. Human and mouse LIF share 79% sequence homology and exhibit cross-species activity, though stem cell differentiation inhibition is mouse-specific.
