Target Information
The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: N-terminal domain, DNA-binding domain, and androgen-binding domain. AR acts as a ligand-activated transcription factor binding testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.
AR signaling is essential for male reproductive organ development and also affects nonreproductive tissues such as muscle, hair follicles, and brain. It regulates MAP kinase activity, and pathway abnormalities are linked to diseases like prostate cancer, Kennedy’s disease, and male infertility. Mutations can cause complete androgen insensitivity (CAIS).
