Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor (>90 kDa) has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. It acts as a ligand-activated transcription factor binding testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

The AR signaling pathway is critical for the development and function of male reproductive organs (prostate, epididymis) and also affects nonreproductive tissues such as muscle, hair follicles, and brain. AR is a phosphoprotein that regulates the MAP kinase pathway. Abnormalities in AR signaling are linked to diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in this gene are associated with complete androgen insensitivity (CAIS).
