Target Information
Nesprin 1과 Nesprin 2는 type II integral membrane protein 계열의 단백질을 암호화하는 두 개의 유전자에 의해 생성됩니다. 이들은 대부분의 조직에서 발현되며, 대체적 mRNA 개시 및 스플라이싱을 통해 두 가지 주요 isoform을 형성합니다. 작은 isoform은 N-말단에서 절단되어 있습니다.
이 단백질들은 다수의 spectrin 반복, bipartite 핵 내 위치 서열, 그리고 보존된 C-말단 단일 transmembrane 도메인을 특징으로 합니다. EGFP 융합 단백질 발현 실험에서 핵막에 위치함이 확인되었으며, perinuclear localization에는 C-말단 TM 도메인이 필수적입니다. Nesprin 1은 평활근 및 골격근 발달 과정에서 발현이 조절되며, myoblast 분화 동안 핵막에서 핵 및 세포질로 재배치됩니다. Nesprin 단백질은 핵 구조 유지에 중요한 역할을 하는 “dystrophins of the nucleus”로 기능할 수 있습니다.
