Target Information
Cyclin D1 (PRAD-1, bcl-1) is a key cell cycle regulator that functions with cdk4 and/or cdk6 to phosphorylate the Rb protein. It is a putative proto-oncogene overexpressed in various human neoplasms including mantle cell lymphomas (MCL). Cyclin D1 regulates protein phosphorylation, mammary gland epithelial cell proliferation, and fat cell differentiation.
The CCND1 gene encoding Cyclin D1 is located on chromosome 11. Cyclin D1 is synthesized during G1 phase, assembling with CDK4 or CDK6 upon growth factor stimulation. These complexes inactivate Rb protein and regulate CDK inhibitors such as p21Cip1 and p27Kip1.
Without stimulation, Cyclin D1 undergoes ubiquitin-mediated degradation, contributing to G1/S phase arrest. It belongs to the conserved cyclin family, functioning as a CDK regulator. Mutations, amplification, and overexpression of Cyclin D1 are frequently observed in tumors and may contribute to tumorigenesis.
Cyclin D1 is a valuable tool for studies on cell cycle biology and cancer-related abnormalities.
