Target Information
Signal-mediated nuclear import and export occur through the nuclear pore complex (NPC), composed of approximately 50 unique nucleoporins. The 98 kDa nucleoporin (NUP98) is generated from a 186 kDa precursor protein via proteolytic cleavage, producing both 98 kDa and 96 kDa nucleoporins localized to the nucleoplasmic side of the NPC. Studies in rats indicate that the 98 kDa nucleoporin functions as a docking site nucleoporin for transport substrates.
The human NUP98 gene can fuse with several genes following chromosomal translocations in acute myelogenous leukemia (AML) and T-cell acute lymphocytic leukemia (T-ALL). It is located within the imprinted gene domain of 11p15.5, a critical tumor-suppressor region. Alterations in this region are linked to Beckwith-Wiedemann syndrome, Wilms tumor, rhabdomyosarcoma, adrenocortical carcinoma, and various cancers including lung, ovarian, and breast cancer. Alternative splicing results in multiple transcript variants, though not all have been fully described.
