| Cellular Localization | Cytoplasm and nucleus. Shuttles between the nucleus and cytoplasm. Translocates into the nucleus upon tyrosine phosphorylation and dimerization in response to activated FGFR signaling. Constitutive nuclear presence is independent of tyrosine phosphorylation. Predominantly cytoplasmic without stimuli. Upon LIF stimulation, accumulates in the nucleus. The BART–ARL2 complex is important for nuclear translocation and retention of STAT3. Background
Defects in STAT3 cause hyperimmunoglobulin E recurrent infection syndrome (AD-HIES, MIM:147060), also known as hyper-IgE or Job syndrome.
This disorder is characterized by immunodeficiency, chronic eczema, recurrent Staphylococcal infections, elevated serum IgE, eosinophilia, coarse facial features, abnormal dentition, joint hyperextensibility, and bone fractures.
Function:
STAT3 acts as a transcription factor binding to IL-6–responsive elements in acute-phase protein gene promoters and is activated by IL31 via IL31RA. It participates in the gp130-mediated signaling pathway.
Post-translational Modifications (PTM):
- Tyrosine phosphorylation in response to IL-6, IL-11, CNTF, LIF, CSF-1, EGF, PDGF, IFN-α, and OSM
- Serine phosphorylation upon DNA damage (likely by ATM or ATR), essential for stable DNA-binding homodimer formation and maximal transcriptional activity.
Structure and Interactions:
- Belongs to the STAT family of transcription factors
- Contains one SH2 domain
- Forms homo- or heterodimers (e.g., with STAT1)
- Interacts with NCOA1, PELP1, SOCS7, STATIP1, HCV core protein, IL23R (in presence of IL23), IL31RA, SIPAR, NLK (via SH2 domain), KPNA4, KPNA5, and TMF1
