Target Information
Post Synaptic Density 95 kDa (PSD-95), also known as synapse associated protein 90 kDa (SAP90), is a membrane-associated protein found in the postsynaptic density of forebrain neurons and certain presynaptic structures in the cerebellum.
PSD-95 contains three PDZ domains, an SH3 domain, and a Guanylate Kinase (GuK) domain. It is involved in clustering proteins such as NMDA receptors and Shaker-type potassium channels at the synaptic membrane in CNS neurons.
There are two main interaction modes between PSD-95 and other proteins:
- NMDA receptors and Shaker-type potassium channels share a C-terminal T/SXV motif that binds to PSD-95 PDZ domains.
- Neuronal nitric oxide synthase (nNOS), lacking the T/SXV motif but containing a PDZ domain, interacts with PSD-95 via PDZ-PDZ association.
