Product Specific Information

The JJ319 monoclonal antibody reacts with the rat CD28 molecule, expressed by thymocytes, mature T cells, and a subset of NK cells. CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2) and serves as a potent costimulator of T cells. Signaling through CD28 augments IL-2 and IL-2 receptor expression as well as cytotoxicity of CD3-activated T cells. No reactivity of JJ319 is observed on mouse T cells.

Applications Tested: Tested by flow cytometric analysis of rat splenocytes. Recommended ≤ 1 µg per test (in 100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test).

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell-specific surface glycoprotein. It belongs to the immunoglobulin superfamily and is constitutively expressed on most peripheral blood T lymphocytes. CD28 acts as a key T cell costimulatory receptor by binding CD80 and CD86 on antigen-presenting cells, preventing T cell anergy or premature apoptosis. In murine lymphoid tissues, all CD4+ and CD8+ T cells express CD28, with expression levels varying by maturation stage.

Diseases associated with CD28 dysfunction include mycosis fungoides and Sezary’s disease.
