Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties and is effective in treating renal, heart, and liver allograft rejection. CD3 is expressed by thymocytes and mature T cells and is critical for TCR complex assembly and activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.2 µg)/test.
Recommended cell number: 10⁵–10⁸ cells/test.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample before combining with cells.
• For single-color controls: use 5 µL per 100 µL cell sample containing 10³–10⁸ cells.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits encoded on chromosome 11. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
